Acute Promyelocytic leukemia (APL or AML-M3) is a subtype of acute myeloid leukemia with distinct clinical and histopathologic features.Genetically, APL cells contain a characteristic translocation fusion gene comprised of the downstream sequences of the retinoic acid receptor alpha gene (RARA) fused to the promoter region and upstream sequences of the promyelocytic leukemia gene (PML).   The fusion gene is designated PML/RARA and is the consequence of the t(15;17)(q22;q21) translocation. Depending on the location of breakpoints within the PML site, intron 6, exon 6 and intron 3, the respective PML-RARa transcript subtypes referred to as long (L or bcr1), variant (V or bcr2) and short (S or bcr3), may be formed. Recent studies have indicated that sensitive monitoring is important because the majority of patients who remain PCR-positive, or become PCR-positive again following treatment, will relapse and likely benefit from early intervention for residual/recurrent disease.

Rely-AMP PML-RARA is an in vitro Real Time polymerase chain reaction assay for the direct, quantitative detection PML-RARA fusion transcript. Housekeeping gene ABL is a reference for relative quantitation.Rely-AMP PML-RARA provides all necessary reagents for RNA extraction, reverse transcription and cDNA detection.
