The leukemogenic fusion gene  transcript E2A-PBX1 results from fusion of the E2A and PBX1 (formerly prl) genes, through the t(1;19)(q23;p13). The t(1;19) is detected in about 5–6% of childhood acute lymphoblastic leukemia and in about 3% of adult acute lymphoblastic leukemia. In the majority of cases the E2A-PBX1 fusion transcript shows a constant junction site of exon 13 (nucleotide 1518) of E2A26 to the second exon (nucleotide 388) of PBX.

A variant fusion transcript is described in about 5–10% of t(1;19)-positive acute lymphoblastic leukemia. It is characterized by a stretch of 27 nucleotides inserted at the usual junction point between nucleotides 1518 and 388 of the E2A and PBX1 genes, respectively.

Rely-AMP E2A-PBX1 is an in vitro Real Time polymerase chain reaction assay for the direct, quantitative detection E2A-PBX1 fusion transcript. Housekeeping gene ABL is a reference for relative quantitation. Rely-AMP E2A-PBX1 provides all necessary reagents for RNA extraction, reverse transcription and cDNA detection.
